You have to slide the flute back and forth across the touch screen to select notes, and blow into the mic to produce them. The mic is really touchy, often doubling notes for no good reason or not producing them at all despite your effort, and you have to be near-PERFECT to proceed. It's really easy to run out of breath in this song, since it's six notes. To make it worse, I play on my 3DS, so I have to hold my hand in a weird way so I don't cover the mic, since it's near the lower right of the touch screen.
