A thief can open it too. The key is there to make sure you still have a chance if you lose all thieves, and it's droppable on an enemy near the door. It would be very hard to lose it before getting to use it. Not sure why the boss started moving, though. Normally he and his pal only start moving after they get attacked. I imagine that opening the door also makes them move, but the fact that the door didn't get opened is kind of the whole crux of this situation.
